PMC-122 is a bispecific monoclonal antibody developed by PharmAbcine that simultaneously targets Programmed Death-Ligand 1 (PD-L1) and Cluster of Differentiation 47 (CD47). By neutralizing both the PD-1/PD-L1 and SIRPα/CD47 immune checkpoint pathways, PMC-122 aims to enhance the anti-tumor immune response through the dual activation of T-cells and macrophages. A key design feature of PMC-122 is its optimized binding affinity, which is engineered to prioritize binding to tumor cells within the tumor microenvironment while minimizing interaction with CD47-expressing red blood cells (RBCs), thereby reducing the risk of hematologic toxicities such as anemia. The drug is currently being investigated as a next-generation immunotherapy for various cancers.
