Overview

PMC-309 is a fully human IgG1 monoclonal antibody developed by PharmAbcine that targets VISTA (V-domain Ig suppressor of T cell activation), a negative immune checkpoint molecule expressed on immunosuppressive cells within the tumor microenvironment. By binding to VISTA, PMC-309 inhibits the function of myeloid-derived suppressor cells (MDSCs), M2 macrophages, and regulatory T cells (Tregs), thereby promoting T cell activation and proliferation. This mechanism leads to enhanced anti-tumor immune responses through increased secretion of pro-inflammatory cytokines such as IFN-gamma, TNF-alpha, and IL-2, as well as the promotion of monocyte differentiation into M1 macrophages. The drug is currently in Phase 1 clinical trials for advanced or metastatic solid tumors and has shown preclinical activity in models relevant to diabetic retinopathy and wet age-related macular degeneration[1][2][3][4][5][6][7][8].
