PMC-309 is a monoclonal IgG1 antibody that specifically targets VISTA (V-domain Ig suppressor of T cell activation), an immune checkpoint molecule expressed on immunosuppressive cells within the tumor microenvironment. By inhibiting VISTA, PMC-309 aims to enhance anti-tumor immune responses. Pembrolizumab is a humanized monoclonal antibody that blocks PD-1 (programmed death receptor 1) on lymphocytes, preventing its interaction with PD-L1/PD-L2 and thereby promoting T-cell mediated anti-tumor activity. The combination of PMC-309 and pembrolizumab is being investigated as a next-generation immuno-oncology therapy for patients with advanced or metastatic solid tumors who may not respond adequately to existing checkpoint inhibitors alone. This combination is currently in Phase 1 clinical trials to assess safety, tolerability, and preliminary efficacy[2][3][4][5][8].
