Overview

PMC-403 is a fully human monoclonal antibody that binds to and activates the Tyrosine-protein kinase receptor Tie2 (TIE2), a key regulator of angiogenesis and vessel maturation in endothelial cells. By activating Tie2, PMC-403 stabilizes pathological, leaky blood vessels and normalizes vascular structure. This mechanism is being investigated for its therapeutic potential in diseases characterized by abnormal vasculature, including neovascular age-related macular degeneration (nAMD), idiopathic systemic capillary leak syndrome (ISCLS/Clarkson disease), diabetic retinopathy, solid tumors, and other vascular-related rare diseases. The drug has received orphan drug designation from the US FDA for ISCLS and is currently in Phase 1 clinical trials for nAMD patients who are unresponsive to anti-VEGF therapies[1][2][3][4][5][6].
