Overview

PMD-336 is a short synthetic peptide therapeutic derived from the active domain of Pigment Epithelium-Derived Factor (PEDF). Developed by Pamdeca, it is designed to reprogram the tumor microenvironment by repolarizing tumor-associated macrophages (TAMs) from an immunosuppressive M2 phenotype to a tumor-suppressive M1 phenotype. This repolarization is characterized by a shift in cytokine secretion, specifically increasing IL-12 and decreasing IL-10, and enhancing macrophage-mediated killing of cancer cells via the TRAIL/DR5 signaling pathway. In preclinical models of ovarian cancer, PMD-336 and its related analog PMD-427 have demonstrated the ability to reduce tumor burden and induce selective apoptosis in malignant cells by modulating extrinsic death cascades involving Fas and FasL.
