Overview

PMD-427 is a short synthetic peptide derived from the active domain of Pigment Epithelium-Derived Factor (PEDF), developed by Pamdeca LLC. It is a first-in-class multi-targeted agent investigated for the treatment of ovarian cancer. PMD-427 functions by reprogramming tumor-associated macrophages (TAMs) from an immunosuppressive M2 phenotype to a tumor-suppressive M1 phenotype, characterized by increased IL-12 and decreased IL-10 expression, as well as attenuated PD-L1 levels. Furthermore, the peptide induces selective apoptosis in ovarian cancer cells by modulating the Fas/FasL extrinsic death cascade and enhancing macrophage-mediated tumor cell killing through the TRAIL/DR5 pathway. Preclinical studies in mouse models have shown that PMD-427 can significantly reduce tumor burden and eradicate ovarian tumors.
