Overview

pmel-1 ACT (Adoptive Cell Therapy) is a form of cancer immunotherapy that utilizes T cells specifically engineered or selected to recognize the pmel-1 (also known as gp100) antigen. This antigen is a lineage-specific differentiation protein involved in melanin synthesis and is highly overexpressed in most human melanomas. The therapy involves the ex vivo expansion of autologous T cells, which are often modified via viral vectors to express a high-affinity T-cell receptor (TCR) specific for a gp100 peptide (such as gp100:209-217) presented by MHC molecules (typically HLA-A*0201). Once re-infused into the patient, these T cells target and eliminate melanoma cells. The "pmel-1" nomenclature is particularly prominent in preclinical research, referring to a widely used TCR transgenic mouse model developed by the National Cancer Institute (NCI) to study CD8+ T cell responses to self-antigens and to optimize ACT protocols, including the use of lymphodepletion and cytokine support like IL-2.
