Pmel-1 adoptive cell transfer (ACT) is a preclinical research model and experimental immunotherapy approach involving the adoptive transfer of CD8+ T cells derived from pmel-1 T-cell receptor (TCR)-transgenic mice. These T cells express a specific TCR (Vα1 and Vβ13) that recognizes the glycoprotein 100 (gp100/pmel-17) epitope, a melanocyte differentiation antigen frequently overexpressed in melanoma. This model is primarily used in the B16 murine melanoma system to evaluate the efficacy of adoptive cell therapies, conditioning regimens (like lymphodepletion), cytokine support (e.g., IL-2), and vaccination strategies. It serves as a foundational platform for studying T-cell persistence, exhaustion, and antitumor activity in a syngeneic immunocompetent setting.
