PMLA-linear-Gd-DOTA NIA is a biodegradable, polymalic acid (PMLA)-based nano imaging agent (NIA) conjugated with linear gadolinium-DOTA (Gd-DOTA) contrast moieties. Developed by researchers at Cedars-Sinai Medical Center, this agent is designed for non-invasive magnetic resonance imaging (MRI) "virtual biopsy" to differentially diagnose primary brain tumors (such as gliomas) and brain metastases. The PMLA polymer scaffold is functionalized with targeting ligands, such as anti-transferrin receptor (TfR) antibody or Angiopep-2 (AP-2) peptide, to facilitate receptor-mediated transcytosis across the blood-brain barrier (BBB). Additionally, it is conjugated with monoclonal antibodies targeting EGFR or HER2 to selectively localize to overexpressing tumor tissues, enabling high-contrast, tumor-specific MRI visualization.
