Overview

PMMB232 is a **synthetic shikonin derivative** with potent anticancer activity, specifically demonstrated against human cervical cancer (HeLa cells). Its mechanism of action involves inducing apoptosis via reduction of mitochondrial membrane potential, promoting PARP cleavage, and downregulating hypoxia-inducible factor 1-alpha (HIF-1α), thereby impairing glycolysis. PMMB232 also upregulates E-cadherin and PDH-E1α while downregulating PDK1. Docking studies suggest a direct binding to HIF-1α[1][3][7]. The compound exhibited a strong antiproliferative effect with an IC50 of 3.25 ± 0.35 μM in vitro and is proposed as a potential anticancer agent targeting tumor cell metabolism and survival[1][3][7].
