PMN267 is a humanized IgG1 monoclonal antibody developed by ProMIS Neurosciences. It is designed to selectively target and bind toxic misfolded aggregates of the TAR DNA-binding protein 43 (TDP-43), which are implicated in the pathogenesis of amyotrophic lateral sclerosis (ALS) and other TDP-43 proteinopathies. The antibody demonstrates high affinity for pathological, misfolded forms of TDP-43 while sparing normal TDP-43, aiming to inhibit cell-to-cell transmission of these toxic aggregates. Preclinical studies have shown that both conventional antibody and intrabody versions of PMN267 can reduce intracellular TDP-43 aggregates and prevent disability in ALS models[5][6][7]. The drug is currently in preclinical development.
