Overview

PMN440 is a therapeutic vaccine in preclinical development by ProMIS Neurosciences. It is designed to target synucleinopathies, including Parkinson's disease, dementia with Lewy bodies, and multiple system atrophy. The vaccine uses computationally-derived conformational B cell epitopes to selectively induce high-affinity antibodies against pathogenic species of alpha-synuclein (toxic oligomers and small soluble seeding fibrils), while sparing normal alpha-synuclein function. This selectivity aims to maximize efficacy and minimize off-target effects by avoiding pan-alpha-synuclein reactivity[6][10]. Preclinical studies have demonstrated that vaccination with these epitopes produces the desired antibody response in animal models[6].
