Overview

PN-1007 is a synthetic glycopolymer that mimics the human natural killer-1 (HNK-1) carbohydrate epitope. It was developed by Polyneuron Pharmaceuticals for the treatment of anti-myelin-associated glycoprotein (MAG) neuropathy, a rare autoimmune disorder of the peripheral nervous system characterized by monoclonal IgM autoantibodies targeting myelin. The drug acts as an immunotherapy and immunosuppressant by selectively binding to and neutralizing circulating anti-MAG IgM autoantibodies, thereby preventing their pathogenic binding to myelin and potentially protecting nerve integrity. Preclinical studies demonstrated that intravenous administration of PN-1007 efficiently removed anti-MAG IgM antibodies in animal models without activating immune cells or inducing cytokine release ex vivo in human samples. Clinical development included phase I/II trials, but these were terminated after initial data failed to support further development[1][2][4][5][6].
