PN-101 is an investigational therapy developed by Paean Biotechnology for the treatment of polymyositis and dermatomyositis. It consists of mitochondria isolated from umbilical cord mesenchymal stem cells and is designed to be transplanted into patients. The mechanism of action involves mitochondrial replacement, aiming to enhance mitochondrial repair and reduce inflammation in muscle tissues. Preclinical studies have shown that PN-101 improves muscle differentiation, increases intracellular ATP content, enhances cell viability, and reduces myositis severity through anti-inflammatory effects. Clinical trials (Phase 1/2a) in patients with refractory polymyositis or dermatomyositis have demonstrated safety and at least minimal clinical improvement compared to baseline[1][2][3][4][5].
