PN-106

PN-106 is a discontinued topical small molecule uridine prodrug that was developed by Wellstat Therapeutics (formerly Pro-Neuron) for the treatment of actinic keratosis (AK). Actinic keratosis is a common precancerous skin lesion caused by chronic exposure to ultraviolet radiation, which leads to DNA damage in keratinocytes. PN-106 was formulated as a topical gel and reached Phase 2 clinical trials (notably NCT00066144), where its safety and efficacy in reducing the number of AK lesions were evaluated. The therapeutic rationale for PN-106 involved the delivery of exogenous uridine to skin cells to support pyrimidine metabolism and potentially enhance DNA repair mechanisms or cellular recovery in sun-damaged tissue. Development of the compound was ultimately halted following the completion of Phase 2 studies.
