PN-2034

PN-2034 (also known as SAR 176975) is a first-in-class oral insulin sensitizer developed for the treatment of type 2 diabetes mellitus. It was discovered by Wellstat Therapeutics and later licensed to Sanofi for global development and commercialization. The drug acts primarily in the liver to restore its capability to respond to insulin, targeting a mechanism distinct from other known insulin sensitizers. Its primary mechanism of action is as an agonist of peroxisome proliferator-activated receptor gamma (PPARγ), which plays a key role in regulating glucose metabolism and improving insulin sensitivity. PN-2034 was considered potentially advantageous over existing therapies due to its novel target and possibly improved side effect profile, particularly regarding weight gain and edema commonly associated with other agents in this class[1][3][5][7].
