Overview

PN-458 is a humanized monoclonal antibody designed as an agonist of the Triggering Receptor Expressed on Myeloid cells 2 (TREM2). Developed by Pionyr Immunotherapeutics as part of its "Myeloid Tuning" platform, PN-458 is intended to stimulate TREM2 signaling on myeloid cells, such as macrophages and microglia. In the context of oncology, TREM2 agonism is hypothesized to reprogram immunosuppressive tumor-associated macrophages (TAMs) into a pro-inflammatory, anti-tumor phenotype (M1-like polarization), thereby enhancing the immune system's ability to attack solid tumors. This mechanism distinguishes it from Pionyr's lead candidate, PY314, which is a TREM2-depleting antibody. Additionally, due to the role of TREM2 in microglial phagocytosis and neuroinflammation, PN-458 has been explored preclinically for neurodegenerative conditions like Alzheimer's disease. PN-458 was part of the pipeline acquired by Gilead Sciences in 2020, but development was discontinued in 2023 following a strategic review of the Pionyr portfolio.
