Drug intelligence / Profile preview
PN-4580 (also referred to as PN-458) is an investigational, wholly owned dual glucagon-like peptide-1 (GLP-1) and gastric inhibitory polypeptide (GIP) receptor agonist peptide being developed by Protagonist Therapeutics for the treatment of obesity. Discovered using Protagonist's proprietary peptide technology platform, PN-4580 is designed to mimic the action of endogenous incretin hormones to promote satiety, delay gastric emptying, and improve glycemic control. The candidate is currently undergoing IND-enabling studies with optionality for both oral (PN-458o) and subcutaneous (PN-458sc) administration. Phase 1 clinical trials are anticipated to initiate in the second half of 2027.
