PN-943 is an orally stable, gastrointestinal-restricted peptide that acts as a selective antagonist of the α4β7 integrin on leukocytes. By binding specifically to this integrin, PN-943 blocks leukocyte trafficking and activation in the gut, thereby inhibiting colon inflammation and reducing signs and symptoms of active ulcerative colitis. The drug was developed as an oral alternative to injectable α4β7 integrin antibody therapies for inflammatory bowel disease. Clinical studies demonstrated dose-dependent pharmacodynamic effects with minimal systemic exposure due to its gut-restricted nature[1][3][5][7].
