Overview

PN23114 is an investigational hepcidin mimetic peptide developed by Protagonist Therapeutics for the treatment of sickle cell disease (SCD). By mimicking the natural hormone hepcidin, PN23114 binds to ferroportin—the only known cellular iron exporter—leading to its internalization and degradation. This process results in iron-restricted erythropoiesis, which reduces the availability of iron for the synthesis of sickle hemoglobin (HbS). Preclinical data presented at the 2023 American Society of Hematology (ASH) Annual Meeting demonstrated that PN23114 can improve hematological parameters and reduce disease-related complications in mouse models of SCD, suggesting its potential as a therapeutic for managing this inherited hemolytic disorder.
