PN277 is an investigational peptide-based immunotherapy developed by Proneuron Biotechnologies for the treatment of neurodegenerative diseases, including Parkinson's disease, spinal cord injury, and stroke. The therapy is based on the concept of "protective autoimmunity," a paradigm developed by Dr. Michal Schwartz which suggests that the immune system can be harnessed to provide neuroprotection and promote tissue repair in the central nervous system (CNS). PN277 is designed to selectively activate beneficial T-cell responses that migrate to sites of neuronal damage. Once at the site of injury, these immune cells are intended to release neurotrophic factors and modulate the local inflammatory environment to support neuronal survival and restoration. While PN277 showed promise in preclinical models and received support from organizations like the Michael J. Fox Foundation, its development ceased following the dissolution of Proneuron Biotechnologies in the mid-2000s.
