PNA-33 is a peptide nucleic acid (PNA) inhibitor designed to target microRNA-33 (miR-33). Developed by researchers at the Yale School of Medicine, it is being investigated for the treatment of pulmonary fibrosis (PF), including idiopathic pulmonary fibrosis (IPF). miR-33 is a key regulator of lipid metabolism and mitochondrial homeostasis in macrophages. By inhibiting miR-33, PNA-33 aims to rebalance macrophage metabolism, restore mitochondrial function, and reduce profibrotic activation. In preclinical models, PNA-33 has demonstrated the ability to improve lung function, reduce collagen accumulation, and outperform current standard-of-care therapies like nintedanib and pirfenidone.
