Overview

PNA1 is a peptide-based therapeutic candidate developed by ProNeurogen, currently in Phase 1 clinical trials for the treatment of vascular dementia, traumatic brain injury (TBI), and post-operative cognitive impairment. As part of ProNeurogen's neurovascular program, PNA1 functions as an agonist of the Mas receptor (MasR), a G protein-coupled receptor that mediates the neuroprotective and anti-inflammatory effects of the alternative renin-angiotensin system (RAS) pathway. By targeting the MasR, PNA1 is designed to improve cognitive function, reduce neuroinflammation, and enhance cerebral blood flow in patients with neurovascular disorders. The drug is a formulation of the endogenous peptide Angiotensin-(1-7), which is known to counteract the vasoconstrictive and pro-inflammatory effects of the classical Angiotensin II/AT1 receptor axis.
