Overview

**PNA5** is a novel glycosylated derivative of angiotensin-(1-7) (Ang-(1-7)) designed as a Mas receptor agonist with enhanced brain penetration and bioavailability compared to the native peptide. It acts by activating the Mas receptor, expressed on neurons, glia, and vascular endothelial cells, to reduce reactive oxygen species (ROS) production, inhibit pro-inflammatory cytokine release, decrease neuroinflammation, improve blood-brain barrier integrity, enhance neurovascular coupling and cerebral blood flow, and restore cognitive function. Developed by researchers at the University of Arizona, including Meredith Hay, and advanced by ProNeurogen, PNA5 has demonstrated efficacy in preclinical mouse models of vascular cognitive impairment and dementia (VCID) induced by heart failure, Parkinson's disease-associated cognitive decline, and traumatic brain injury, reversing cognitive deficits without affecting cardiac function.[1][2][3][6][9][10]
