Overview

pNaKtide is a synthetic peptide that acts as a selective inhibitor of the Na/K-ATPase/Src receptor complex, antagonizing ouabain-induced protein kinase signaling and the amplification of reactive oxygen species (ROS). Mechanistically, it targets the Src kinase pool associated with Na/K-ATPase, suppressing downstream signaling including the ERK1/2 pathway. Preclinical studies demonstrate that pNaKtide reduces oxidative stress, adipogenesis, and insulin resistance, and attenuates obesity, hepatic steatosis (NASH), fibrosis, atherosclerosis, and uremic cardiomyopathy in various murine models. Its primary mechanism is interruption of the Na/K-ATPase–driven ROS amplification loop, implicating it as a potential candidate for treatment of obesity, metabolic syndrome, steatohepatitis, and related cardiovascular and renal complications[1][2][3][4][5][7][8].
