Overview

PNB-291 is a small molecule pan-cholecystokinin (CCK) receptor antagonist developed by PNB Vesper Life Sciences. It functions as a mixed antagonist targeting multiple CCK receptor subtypes, including CCK-A and CCK-B (gastrin) receptors, which are implicated in the growth and progression of various tumors. The drug is primarily being developed for the treatment of brain cancer (glioma), with secondary indications in lung and liver cancers. Preclinical studies have demonstrated that PNB-291 inhibits brain cancer cell lines, such as U373MG, at low nanomolar concentrations and significantly reduces tumor volume in human U87 xenograft models following oral administration. The compound has completed preclinical development and is positioned for Investigational New Drug (IND) regulatory filings.
