Drug intelligence / Profile preview
PNC-28 is a synthetic, p53-derived peptide designed to selectively target and destroy cancer cells. It consists of the human double minute 2 (HDM-2/MDM-2) binding domain of the p53 tumor suppressor protein (residues 17–26) fused at its C-terminus to a cell-penetrating penetratin sequence. Unlike native p53, which acts as a transcription factor in the nucleus to induce apoptosis, PNC-28 targets HDM-2 expressed aberrantly on the cell membranes of cancer cells. Upon binding to membrane-bound HDM-2, the peptide undergoes a conformational change that induces transmembrane pore formation (poration), leading to rapid cell lysis and tumor cell necrosis in a p53-independent manner. Sparing normal, untransformed cells, PNC-28 has been investigated preclinically for the treatment of various solid tumors, particularly pancreatic cancer.
