Overview

pneumococcal polysaccharide serotype 10A conjugated to CRM197 adsorbed on aluminium phosphate is a monovalent vaccine candidate developed by the Rijksinstituut voor Volksgezondheid en Milieu (RIVM) for the prevention of pneumococcal infections caused by Streptococcus pneumoniae serotype 10A. The vaccine utilizes a conjugate technology where the purified capsular polysaccharide of serotype 10A is covalently linked to CRM197, a non-toxic mutant of the diphtheria toxin. This conjugation converts the polysaccharide from a T-cell independent antigen to a T-cell dependent antigen, thereby inducing a more robust and long-lasting immune response, including immunological memory, which is particularly effective in infants and young children. The formulation is adsorbed on aluminium phosphate, which acts as an adjuvant to enhance the overall immunogenicity of the vaccine. This specific conjugate is often developed as a component for inclusion in higher-valency multivalent pneumococcal conjugate vaccines (PCVs).
