Overview

Pneumococcal polysaccharide serotype 14 CRM197 conjugate adsorbed on aluminium phosphate is a monovalent vaccine component developed by the Rijksinstituut voor Volksgezondheid en Milieu (RIVM) in the Netherlands. It consists of the purified capsular polysaccharide of *Streptococcus pneumoniae* serotype 14, which is chemically conjugated to the CRM197 carrier protein—a non-toxic, recombinant mutant of the diphtheria toxin. The conjugate is adsorbed onto an aluminium phosphate adjuvant to enhance its immunogenicity. This formulation was developed as part of RIVM's initiative to create a cost-effective 10-valent pneumococcal conjugate vaccine (PCV10) for global health use. By conjugating the polysaccharide to a protein carrier, the vaccine induces a T-cell dependent immune response, which is crucial for providing protection and establishing immunological memory in infants and young children, a population where plain polysaccharide vaccines are ineffective. Serotype 14 is historically one of the most prevalent serotypes causing invasive pneumococcal disease worldwide.
