Overview

The pneumococcal polysaccharide serotype 19A CRM197 conjugate vaccine adsorbed on aluminium phosphate is a monovalent vaccine candidate developed by the Rijksinstituut voor Volksgezondheid en Milieu (RIVM) and Intravacc. It is specifically designed to provide immunity against Streptococcus pneumoniae serotype 19A, a major cause of invasive pneumococcal disease that emerged as a significant pathogen following the introduction of early multivalent vaccines that did not cover this serotype. The vaccine utilizes a conjugation technology where the 19A capsular polysaccharide is covalently bound to CRM197, a non-toxic recombinant mutant of the diphtheria toxin. This conjugation converts the polysaccharide from a T-cell independent to a T-cell dependent antigen, thereby inducing a more robust immune response and immunological memory, particularly in infants. The formulation is adsorbed onto aluminium phosphate, which serves as an adjuvant to further enhance the humoral immune response.
