Pneumococcal polysaccharide serotype 4 conjugated to CRM197 adsorbed on aluminium phosphate is a monovalent vaccine component developed by the Rijksinstituut voor Volksgezondheid en Milieu (RIVM). It consists of the capsular polysaccharide of Streptococcus pneumoniae serotype 4 covalently linked to CRM197, a non-toxic recombinant mutant of the diphtheria toxin. This conjugation converts the T-cell independent polysaccharide antigen into a T-cell dependent antigen, which significantly enhances immunogenicity and induces immunological memory, particularly in infants and young children. The conjugate is adsorbed onto an aluminium phosphate adjuvant to further stimulate the innate and adaptive immune response. This specific conjugate technology was developed by RIVM as part of a decavalent pneumococcal conjugate vaccine (PCV10) project, which was subsequently transferred to the Serum Institute of India for global production and distribution under the brand name PNEUMOSIL.
