Overview

This is a monovalent pneumococcal conjugate vaccine component consisting of the capsular polysaccharide of *Streptococcus pneumoniae* serotype 5 covalently linked to the CRM197 carrier protein. CRM197 is a non-toxic recombinant variant of the diphtheria toxin that serves to convert the polysaccharide into a T-cell dependent antigen, thereby inducing a more robust and long-lasting immune response, particularly in infants. The conjugate is adsorbed onto an aluminium phosphate adjuvant to further enhance immunogenicity. Developed by the Rijksinstituut voor Volksgezondheid en Milieu (RIVM) in the Netherlands, this technology was specifically designed for transfer to manufacturers in developing countries to facilitate the production of affordable multivalent pneumococcal conjugate vaccines (PCVs). The technology was successfully transferred to the Serum Institute of India for the production of Pneumosil, a 10-valent vaccine that includes this serotype 5 conjugate.
