Overview

Pneumococcal polysaccharide serotype 6A conjugated to CRM197 adsorbed on aluminium phosphate is a monovalent vaccine conjugate component developed by the Rijksinstituut voor Volksgezondheid en Milieu (RIVM). It consists of the capsular polysaccharide of *Streptococcus pneumoniae* serotype 6A covalently linked to CRM197, a non-toxic mutant of the diphtheria toxin. This conjugation process converts the polysaccharide from a T-cell independent to a T-cell dependent antigen, which significantly enhances immunogenicity and induces immunological memory, particularly in infants. The conjugate is adsorbed onto an aluminium phosphate adjuvant to further stimulate the immune response. This specific conjugate technology was developed by RIVM and transferred to manufacturers like the Serum Institute of India for use in multivalent pneumococcal conjugate vaccines (PCVs) such as Pneumosil, which are designed to prevent invasive pneumococcal diseases including pneumonia, meningitis, and sepsis.
