pNGVL4a-CRTE6E7L2 is a therapeutic DNA vaccine developed by Johns Hopkins University in collaboration with Ichor Medical Systems for the treatment of HPV16-positive high-grade cervical intraepithelial neoplasia (CIN 2/3). The vaccine utilizes the pNGVL4a plasmid vector to encode a fusion protein comprising calreticulin (CRT) linked to the HPV16 E6 and E7 oncoproteins and the L2 minor capsid protein. This construct is designed to enhance the presentation of HPV antigens to the immune system, specifically inducing potent HPV-specific CD8+ T cell responses against the E6 and E7 oncoproteins and generating neutralizing antibodies against the L2 protein. The vaccine is administered intramuscularly using the TriGrid Electroporation Device, which significantly improves cellular uptake of the DNA and subsequent immune activation compared to conventional injection methods.
