Overview

pNGVL4aCRTE6E7L2 + TA-CIN is an investigational combination immunotherapy developed for the treatment of persistent Human Papillomavirus type 16 (HPV16) infections and associated cervical abnormalities. The regimen consists of two components: pNGVL4aCRTE6E7L2, a naked DNA plasmid vaccine encoding a fusion protein of calreticulin (CRT) linked to HPV16 E6, E7, and L2 antigens; and TA-CIN, a recombinant fusion protein also containing HPV16 L2, E6, and E7. The inclusion of calreticulin in the DNA construct is intended to enhance the MHC class I presentation of the HPV antigens, thereby boosting the CD8+ T-cell response. TA-CIN serves as a protein boost to further stimulate both cellular and humoral immunity. This prime-boost strategy is primarily targeted at patients with HPV16-positive atypical squamous cells of undetermined significance (ASC-US) or low-grade squamous intraepithelial lesions (LSIL) to promote viral clearance and lesion regression.
