PNK-CAR38 is an investigational **allogeneic CAR-NK cell therapy** developed by **Celularity** for hematologic malignancies, particularly **CD38-positive lymphoma** and **multiple myeloma**. The product consists of **human placental CD34-positive hematopoietic stem cell-derived natural killer cells** engineered with a **second-generation anti-CD38 chimeric antigen receptor** containing a CD38-binding scFv linked to **CD28** and **CD3 zeta** signaling domains, then expanded and differentiated into NK cells. Mechanistically, it is designed to direct cytotoxic NK-cell activity against **CD38-expressing malignant cells** as an off-the-shelf cell therapy platform. Preclinical reports indicate enhanced antitumor activity against CD38-positive lymphoma and myeloma cell lines without clear on-target off-tumor cytotoxicity against healthy CD38-positive cells.
