Overview

**PNP-DOX** is a preclinical, PLZ4-targeted nanoporphyrin formulation carrying the anthracycline chemotherapy drug doxorubicin for bladder cancer. It is assembled from pyropheophorbide a-containing and PLZ4-conjugated telodendrimers; PLZ4 binds integrin alpha-V/beta-3 on bladder cancer cells, supporting selective tumor localization. Upon near-infrared illumination, the nanoporphyrin component supports fluorescence-guided photodynamic diagnosis, photodynamic therapy, and photothermal therapy, while released doxorubicin produces chemotherapy through DNA intercalation and topoisomerase II inhibition. The formulation showed selective bladder-tumor accumulation and antitumor activity in orthotopic patient-derived xenograft models, but no clinical development has been identified.
