Overview

PNQ-154 is a small molecule, reversible, non-covalent inhibitor of Bruton's tyrosine kinase (BTK) being developed by Impetis Biosciences. Unlike first-generation covalent BTK inhibitors such as ibrutinib, PNQ-154 is designed to maintain activity in the presence of resistance mutations, such as the C481S mutation, which frequently occur in patients with B-cell malignancies. The drug is intended for oral administration and is currently in the IND-enabling stage for the treatment of ibrutinib-resistant cancers, including diffuse large B-cell lymphoma (DLBCL).
