PNQ-370 is a small molecule antagonist of the A2A adenosine receptor (A2AR) originally developed by Impetis Biosciences and licensed to Tarus Therapeutics (a Roivant Sciences company). The compound is designed to inhibit adenosine-mediated immunosuppression within the tumor microenvironment, where high levels of adenosine typically suppress the activity of T cells and natural killer cells. By blocking A2AR, PNQ-370 aims to restore and enhance the anti-tumor immune response. It is being investigated for the treatment of lung cancer and various other solid tumors. Additionally, due to the role of A2A receptors in modulating dopaminergic signaling, PNQ-370 is being explored for central nervous system (CNS) indications, including Parkinson's disease and cognitive disorders. As of the latest reports, the drug is in the IND-enabling phase of development.
