PNT001

PNT001 is a humanized full-length IgG4 S228P monoclonal antibody that selectively binds the cis conformation of the phosphorylated Thr231-Pro232 motif in human tau protein (cis-pT231 tau), with high affinity and specificity. This pathological form of tau is implicated in neurodegenerative diseases, including traumatic brain injury, chronic traumatic encephalopathy, vascular dementia, and Alzheimer’s disease. By targeting cis-pT231 tau, PNT001 aims to inhibit aggregation and spread of toxic tau species that drive neuronal death and cognitive decline. Preclinical studies have shown efficacy in models of tauopathy, and Phase 1 clinical trials demonstrated safety and tolerability at doses expected to be therapeutic[1][2][3][5][6].
