PNT2001 is a next-generation, small-molecule radiopharmaceutical targeting prostate-specific membrane antigen (PSMA), developed by Scintomics and licensed to Point Biopharma (now a subsidiary of Eli Lilly). Designed for the treatment of metastatic castration-resistant prostate cancer (mCRPC), PNT2001 utilizes a high-affinity ligand that can be labeled with either the beta-emitter Lutetium-177 (177Lu) or the alpha-emitter Actinium-225 (225Ac). This theranostic-ready molecule is engineered to provide superior tumor-to-background ratios compared to first-generation PSMA-targeted agents, potentially improving efficacy while reducing toxicity to healthy tissues such as the salivary glands and kidneys. The program is currently in late-stage clinical development for prostate cancer, representing a follow-on or optimized alternative to earlier PSMA-targeted radioligands.
