Overview

PNU-159682 is a highly potent secondary metabolite of nemorubicin, belonging to the anthracycline class of natural products. It has the molecular formula C32H35NO13 and a molecular weight of 641.62. It is soluble in DMSO (≥100 mg/mL or 155.86 mM) but has poor water solubility (<0.1 mg/mL). It was identified as a bioactivation product of nemorubicin (MMDX) generated by CYP3A4 in human liver. PNU-159682 functions as a DNA topoisomerase II inhibitor and intercalates between CG base pairs of DNA, forming highly stable complexes. It is remarkably more cytotoxic than its parent compound nemorubicin and doxorubicin in vitro. Due to its extreme potency, it is being explored as a promising candidate, particularly as a payload for antibody-drug conjugates (ADCs) and targeted formulations like nanocells.
