Overview

PNU-93914 (also known as LEP-ETU or LipoTaxen) is a liposomal nanoparticle formulation of the chemotherapeutic agent paclitaxel. Developed to improve the therapeutic index and reduce the toxicities associated with conventional paclitaxel (which requires Cremophor EL), this formulation encapsulates the drug within liposomes. As a mitotic inhibitor and antimicrotubule agent, it works by binding to tubulin, stabilizing microtubules, and inhibiting cell division, ultimately leading to apoptosis. It has been primarily investigated by Memorial Sloan Kettering Cancer Center in Phase II clinical trials for the treatment of locally advanced or metastatic esophageal cancer in patients who have previously failed chemotherapy.
