Overview

PNU153429 is a **tetrasulfonated derivative of distamycin A** under investigation as an antiangiogenic small molecule with antitumor and antimetastatic properties. It has been tested primarily in preclinical models, notably for its ability to inhibit angiogenesis and metastasis in Kaposi’s sarcoma-like tumor models induced by extracellular HIV-Tat protein. Its mechanism of action is thought to involve inhibition of angiogenesis and interference with HIV-Tat activity. It has also been evaluated for potential use in rheumatoid arthritis and as a chemotherapy adjunct in bladder cancer. Preclinical studies showed that PNU153429 delayed tumor growth and reduced metastases in mouse xenograft models, with no notable toxicity.
