PNV-5030

PNV-5030 is a **highly selective adenosine A1 receptor antagonist small molecule** developed by Purnovate, a subsidiary of Adial Pharmaceuticals, for the treatment of pain. Preclinical data show significant pain-reducing effects compared to placebo and acetaminophen, with the potential to reduce or eliminate opioid use when administered alone or in combination with morphine. PNV-5030 is notable for its over 1000-fold selectivity over the adenosine A1 receptor and possesses enhanced water solubility (over 50 times greater than similar compounds), addressing prior limitations of this drug class. While current development is focused on pain (both acute and chronic), the adenosine platform is also being considered for broader indications such as inflammatory diseases, diabetes, asthma, cancer, Parkinson's disease, and infectious diseases associated with cytokine storms (including COVID-19). PNV-5030 had not yet entered human trials as of the most recent reports but was nearing first-in-human studies.[1][3][4][7][13]
