Overview

Pocapavir (V-073) is an investigational small molecule antiviral drug that functions as a capsid inhibitor. It is primarily being developed for the treatment of poliovirus infections and other enteroviral diseases, including those caused by Coxsackievirus B3. By binding to the hydrophobic pocket of the viral capsid protein VP1, pocapavir prevents the conformational changes necessary for viral uncoating and the subsequent release of viral RNA into the host cell cytoplasm. The drug has been employed in compassionate use settings for life-threatening neonatal enteroviral sepsis and is being evaluated as part of the Global Polio Eradication Initiative, particularly for immunodeficient individuals who chronically excrete vaccine-derived polioviruses.
