Overview

**PODO447** is a monoclonal antibody that specifically targets a tumor-restricted glycoepitope on podocalyxin (Podxl), consisting of a non-sialylated core 1 O-linked glycan (T-antigen) in the context of the Podxl peptide backbone. This epitope is expressed on over 60-65% of high-grade serous ovarian carcinomas and various other Podxl-expressing tumors including pancreatic, glioblastoma, and leukemia, but not on normal adult tissues such as kidney podocytes or vascular endothelia. Developed by researchers at the University of British Columbia, PODO447 lacks inherent blocking activity against Podxl function but internalizes efficiently and, when conjugated to cytotoxic payloads like monomethyl auristatin E (MMAE) or Vedotin, forms highly effective antibody-drug conjugates (ADCs) that selectively kill tumor cells in vitro and demonstrate potent efficacy against human ovarian and pancreatic tumor xenografts in mouse models. It also shows modest antibody-dependent cellular cytotoxicity (ADCC) and serves as a biomarker for immune-cold tumors with poor CD8+ T-cell and CD20+ B-cell infiltration.[1][2][3][5][6][9]
