Podoplanin (PDPN), also known as Aggrus or T1-alpha, is a type I transmembrane sialoglycoprotein that is highly expressed on lymphatic endothelial cells and upregulated in various human cancers, including glioblastoma, malignant mesothelioma, and squamous cell carcinomas. Podoplanin antibodies are designed to bind to the extracellular domain of PDPN, particularly the platelet aggregation-stimulating (PLAG) domains. By blocking the interaction between PDPN and the C-type lectin-like receptor 2 (CLEC-2) on platelets, these antibodies can inhibit tumor cell-induced platelet aggregation and hematogenous metastasis. Beyond therapeutic applications, podoplanin antibodies are widely used as diagnostic tools in flow cytometry and immunohistochemistry to identify lymphatic vessel invasion and to distinguish specific malignancies, such as Acute Promyelocytic Leukemia (APL), from other forms of leukemia.
