PODS BMP-2 is a sustained-release crystalline protein formulation in which human bone morphogenetic protein-2 (BMP-2) is co-crystallized with the polyhedrin scaffold protein using the PODS (Polyhedrin Delivery System) technology. In this system, BMP-2 is expressed in insect cells together with polyhedrin, which self-assembles into microcrystals that entrap the nascent, natively folded BMP-2, protecting it from proteolytic degradation and enabling prolonged, protease-dependent release of active growth factor in vitro and in vivo.[1][4] Once liberated from the crystals, BMP-2 functions as a dimeric member of the TGF-β superfamily that binds type I and type II BMP receptors and signals via SMAD pathways to potently induce osteoblast and chondrocyte differentiation and promote bone and cartilage formation, with additional roles in cardiac development and epicardial cell migration.[1][4][6] PODS BMP-2 is currently supplied for research use, including applications in cartilage repair and osteoarthritis models where it has been shown to enhance chondrocyte proliferation, extracellular matrix production, and cartilage regeneration compared with conventional soluble BMP-2.
