Overview

POH1 siRNA is a small interfering RNA designed to silence the expression of the POH1 gene (also known as PSMD14 or Rpn11). POH1 is a highly conserved deubiquitinating enzyme (DUE) that serves as a critical component of the 19S proteasome lid, where it facilitates protein degradation by removing ubiquitin chains from substrates. In oncology research, POH1 knockdown has been shown to inhibit tumor cell proliferation and induce apoptosis via the mitochondrial pathway, often involving the stabilization of p53 and Bim. While primarily used as a research reagent in laboratory settings, preclinical studies have explored its therapeutic potential through intratumoral delivery in models of hepatocellular carcinoma, esophageal cancer, and colorectal cancer.
